چکیده

Nakamura, Hakobu (Konan University, Kobe, Japan). Acriflavine-binding capacity of Escherichia coli in relation to acriflavine sensitivity and metabolic activity. J. Bacteriol. 92:1447-1452. 1966.-Inheritance of the acriflavine resistance gene by an acriflavine-sensitive strain of Escherichia coli K-12 resulted in a reduction in the cellular accumulation of acriflavine and other basic dyes, but had no effect on the accumulation of acid dyes. In both acriflavine-resistant and -sensitive strains, the cooling of cells from 37 to 0 C increased acriflavine accumulation. This increased accumulation was released from the cells after restoration to 37 C. Acriflavine accumulation was increased by carbon shortage and by metabolic disturbance caused by potassium cyanide, arsenite, puromycin, chloramphenicol, 2-thiouracil, and 8-azaguanine. The functional relation of acrifiavine accumulation to the acriflavine concentration of the medium suggests that adsorption is involved in the accumulation.

In Vitro Study of Acriflavine Interaction with Horseradish Peroxidase C

Acriflavine (3,6-diaminoacridine) is an anticeptic drug developed in 1912. Previous research has focused on investigation of the intercalating features of acriflavine, but little is known about its interaction with proteins. Drug-receptor interaction is of major interest in clinical science. The aim of the present study was to evaluate the ability of acriflavine to induce alterations in conform...

Acridine binding by Escherichia coli: pH dependency and strain differences.

Acridine dye binding by cells of Escherichia coli has been characterized in terms of a number of parameters. There is a temperature-dependent, readily reversible binding of acriflavine which occurs to a greater extent with acridine-sensitive mutants of E. coli K-12 than with wild-type E. coli B or K-12. There is an essentially irreversible internal binding of acriflavine which occurs when the c...

EfrAB, an ABC multidrug efflux pump in Enterococcus faecalis.

A DNA fragment responsible for resistance to antimicrobial agents was cloned from the chromosomal DNA of Enterococcus faecalis ATCC 29212 by using drug-hypersensitive mutant Escherichia coli KAM32 as a host cell. Cells of E. coli KAM32 harboring a recombinant plasmid (pAEF82) carrying the DNA fragment became resistant to many structurally unrelated antimicrobial agents, such as norfloxacin, cip...

VmrA, a member of a novel class of Na(+)-coupled multidrug efflux pumps from Vibrio parahaemolyticus.

Gene vmrA, cloned from Vibrio parahaemolyticus, made Escherichia coli resistant to 4',6-diamino-2-phenylindol, tetraphenylphosphonium chloride, acriflavine, and ethidium bromide. VmrA belongs to the DinF branch of MATE family efflux transporters. VmrA catalyzed acriflavine efflux and showed Na(+)/drug transporter activity because the addition of tetraphenylphosphonium to Na(+)-loaded cells caus...
